Principles of the Reaction
Cupric ions in an alkaline solution form a violet colored complex with the carbonyl and amino groups of peptide linkages in protein. The absorbance of the complex is quantitated at 546 nm and is directly proportional to the total protein concentration in the sample.

The buffer used is optimized for the solubilization of intervenously infused dextran plasma expanders so as to eliminate this common source of interference in certain biuret reagents.
Expected Values
The adult normal range for total protein is reported to be 60-85 g/L (6.0-8.5 g/dl).
Special Performance Characteristics
- The Synermed® total protein methodology was correlated to a widely accepted biuret method. The calculated linear regression on 44 samples ranging from approximately 15 to 100 g/L (1.5-10 g/dl) with Synermed on the Y-axis was: Y = 1.01X - 0.9 g/L (0.09 g/dl) with a correlation coefficient of 0.999.
- The sensitivity of the procedure is such that an absorbance change of 0.001 will detect as little as 0.36 g/L (0.036 g/dl) of total protein.
- The average within-run precision of the method was determined by assaying 30 samples of quality control material at two levels of total protein concentration which provided the following results:
Mean SD CV 43.7 g/L (4.4 g/dl) 0.26 (.026) 0.6% 65.6 g/L (6.6 g/dl) 0.26 (.026) 0.4% - The run to run reproducibility of the method as applied to an automated analyzer was determined from the values obtained by 5 replicate analyses of quality control material assayed over a 22 batch period. The following results were obtained:
